Proguanil

An antimalarial taken as a combination tablet with atovaquone to prevent and treat malaria.

Clinical information

Common uses

  • preventing falciparum malaria, including chloroquine-resistant areas
  • treating uncomplicated falciparum malaria

Common side effects

  • headache
  • diarrhoea
  • abdominal pain
  • feeling sick and vomiting
  • mouth ulcers

Cautions

  • not for prevention with severe kidney impairment
  • liver problems including hepatitis reported
  • severe skin reactions can occur — stop immediately
  • not suitable for severe or cerebral malaria

Interactions

  • rifampin or rifabutin — not recommended
  • warfarin (blood thinner) — effect may be increased
  • tetracycline — monitor for reduced antimalarial effect

Pregnancy: only if prescribed — keep taking folate supplements
