Lenalidomide

Lenalidomide (Revlimid) is a thalidomide-related capsule for multiple myeloma and some blood cancers.

Clinical information

Common uses

  • multiple myeloma
  • anaemia in del(5q) myelodysplastic syndromes
  • relapsed mantle cell lymphoma
  • follicular or marginal zone lymphoma (with rituximab)

Common side effects

  • diarrhoea
  • tiredness
  • anaemia
  • constipation
  • low white blood cells

Cautions

  • causes severe birth defects — strict pregnancy prevention required
  • can cause serious drops in blood counts — regular blood tests
  • raised risk of blood clots, heart attack and stroke
  • only available through a restricted access programme

Interactions

  • digoxin (levels rise; monitor)
  • erythropoietin-stimulating agents (higher clot risk)
  • oestrogen-containing therapies (higher clot risk)
  • pembrolizumab (increased deaths in myeloma trials)

Pregnancy: must never be used — causes severe birth defects
