Crizotinib

A targeted cancer medicine (kinase inhibitor) for ALK- or ROS1-positive cancers.

Clinical information

Common uses

  • ALK/ROS1-positive metastatic non-small cell lung cancer
  • ALK-positive anaplastic large cell lymphoma (children, young adults)
  • ALK-positive inflammatory myofibroblastic tumour

Common side effects

  • vision problems
  • nausea, vomiting and diarrhoea
  • swelling (oedema) and constipation
  • tiredness and reduced appetite
  • dizziness and nerve symptoms

Cautions

  • can cause fatal liver damage — regular liver tests
  • lung inflammation (pneumonitis) — stop permanently if it occurs
  • heart rhythm changes and slow heartbeat
  • can harm an unborn baby — use contraception

Interactions

  • strong CYP3A inhibitors — avoid
  • strong CYP3A inducers — avoid
  • sensitive CYP3A substrate drugs — avoid

Pregnancy: avoid — can harm the baby
