Bosentan

A tablet for pulmonary arterial hypertension (high pressure in lung arteries).

Clinical information

Common uses

  • pulmonary arterial hypertension in adults
  • pulmonary arterial hypertension in children 3 years and older

Common side effects

  • respiratory tract infection
  • anaemia (low red blood cells)

Cautions

  • can cause serious liver injury — monthly liver tests required
  • must not be used in pregnancy
  • only available through a restricted programme (REMS)
  • fluid retention and low haemoglobin can occur

Interactions

  • ciclosporin — must not be combined
  • glyburide — must not be combined
  • hormonal contraceptives — made less effective
  • CYP2C9 inhibitor plus strong/moderate CYP3A inhibitor — not recommended
  • drugs broken down by CYP3A or CYP2C9 — levels reduced

Pregnancy: must not be used — may cause birth defects; use contraception
