Common uses
- pulmonary arterial hypertension in adults
- pulmonary arterial hypertension in children 3 years and older
Common side effects
- respiratory tract infection
- anaemia (low red blood cells)
Cautions
- can cause serious liver injury — monthly liver tests required
- must not be used in pregnancy
- only available through a restricted programme (REMS)
- fluid retention and low haemoglobin can occur
Interactions
- ciclosporin — must not be combined
- glyburide — must not be combined
- hormonal contraceptives — made less effective
- CYP2C9 inhibitor plus strong/moderate CYP3A inhibitor — not recommended
- drugs broken down by CYP3A or CYP2C9 — levels reduced
Pregnancy: must not be used — may cause birth defects; use contraception
