Abemaciclib

A targeted kinase-inhibitor tablet used to treat certain breast cancers.

Clinical information

Common uses

  • HR-positive, HER2-negative early breast cancer (with endocrine therapy)
  • advanced or metastatic HR-positive, HER2-negative breast cancer

Common side effects

  • diarrhoea
  • low white blood cell counts
  • feeling sick and vomiting
  • tiredness
  • loss of appetite

Cautions

  • severe diarrhoea with dehydration can occur
  • lung inflammation (pneumonitis) can be severe or fatal
  • liver problems — regular blood tests needed
  • can harm an unborn baby

Interactions

  • ketoconazole (avoid)
  • other strong/moderate CYP3A inhibitors (dose reduction)
  • strong/moderate CYP3A inducers (avoid)
